       Synthesis and Evaluation of New Chemical Entities with special reference to heterocyclic moieties :

 

Brief description of work done:

 

Various novel analogs of imidazoles, 2-methylimidazoles and 2-methyl-5-nitroimidazoles have been synthesized, characterized and evaluated for their antibacterial, antifungal, antidiarrheal, anticonvulsant and miscellaneous other activities.1- substituted imidazoles, 2-methylimidazoles and 2-methyl-5-nitroimidazoles have showed interesting activities in the field of antimycobacterial , anti fungal , anticonvulsant and  antihypertensive research.

The benzimidazole scaffold has also been used for synthesis of novel mercaptobenzimidazoles attached to the quinazoline moiety which have been evaluated for their antiulcer activity. It is worth noting that some of the novel benzimidazoles have shown potent antiulcer activities.

Similarly, novel quinazolines have been synthesized which have shown very high activities against influenza virus and herpes viruses.

   

·         Insiilco  Design, Synthesis and Evaluation of antiHIV  agents.

 

Brief description of work done:

 

Three dimensional QSAR as well as docking studies of various HIV-1- RT inhibitors, particularly non nucleoside reverse transcriptase inhibitors have been carried out. Based on the QSAR and modelling studies , various heterocyclic moieties for e.g. tetrahydroquinolines and tetrahydroisoquinolines, N-aminoimidazoles, thiazolidinediones, tetrazoles, , bernzotriazoles, and benzimidazoles have been used as scaffolds for the design and synthesis of novel analogs of these heterocycles which have been evaluated for HIV-1 -RT inhibitory activities along with antimicrobial activities to develop antiHIV agents with broad spectrum chemotherapeutic properties. Some of the N- aminoimidazole analogs have shown interesting activities against the K103N and Y181C resistant strains of HIV.

Encouraging results have also been found with teterahydroquinolines   , tetrahydroisoquinolines and imidazoles as as dual inhibitors, exhibiting both antiHIV and antibacterial properties. Work is also being carried out in the field of HIV integrase inhibitors by modelling, synthesis and evaluation of novel diketo acids.

 

·         Molecular Modeling and Cheminformatics.

 

Brief description of work done:

 

QSAR , modelling  and ADMET studies using various modelling software viz., SYBYL/CoMFA and COMSIA, Schrodinger suite, FlexX, Scigress Explorer Ultra and ADME Works Model Builder for various azoles and tetrahydroquinolines have been carried out to develop predictive and robust models leading to the design of newer azoles with potential antifungal and antiHIV activities.

 

Level of already achieved success: The Principal Investigator of this proposal has been working on ‘Imidazole analogs’ since 1994. She has several collaborations with different international research laboratories and  groups working in the field of anti HIV agents like  Rega Institute for Medical Research, Belgium ; Institute of Molecular Genetics , National Research Council, Italy ; Division of Infectious Diseases, University of Pittsburgh, School of Medicine, USA; TAACF ,Southern Research Institute ,Birmingham, and with Developmental Therapeutics Program, NCI, NIH, USA. On a national level she has collaboration with Indian Institute of Natural Resins and Gums, Namkom, Ranchi etc. Her experience about different experiments, e.g,. synthesis  of NCEs and molecular modelling etc. will be found useful for the proposed works. The PI has completed three research projects; one AICTE RPS project on ‘DAMNI analogs’; one UGC Minor Research project on “Synthesis, evaluation and molecular modelling of imidazole analogs” and one UGC Major Research Project on “In Silico Design, Synthesis and Evaluation of newer azoles as HIV-1-RT inhibitors. The PI has found very interesting results particularly with imidazole analogs against HIV-1 RT inhibitors.
